Purpose: Juniper (Juniperus procera) is a common forest tree species in Saudi Arabia. The decline in many populations of J. procera in Saudi Arabia is mainly due to seed dormancy and loss of natural regeneration. This study assessed the effects of chemical and hormonal treatments on seed germination and seedling growth in juniper plants. Methods: The seeds were subjected to either chemical scarification with 90% sulfuric acid and 20% acetic acid for 6 min or hormonal treatment by seed soaking in two concentrations (50 and 100 ppm) of three growth regulators, namely, indole acetic acid (IAA), gibberellins (GA3), and kinetin, for 72 h. A control group without any seed treatment was also prepared. The experiments were performed in an incubator maintained at room temperature and under a light and dark period of 12 h for 6 w. The germinated seeds for each treatment were counted and removed from the dishes. The selected germinated seeds from different treatments were planted in a greenhouse and irrigated with tap water for another 6 weeks. The hormone-treated seedlings were sprayed with their corresponding hormone concentrations 1 w after planting. Results: The highest percentage of seed germination was significantly recorded after seed soaking in 50 ppm GA3, whereas treatment with IAA (100 ppm) resulted in the best seedling growth. Seedlings treated with the three phytohormones showed a significant increase in photosynthetic pigments, total soluble sugars, proteins, percentage of oil, IAA, GA3, and kinetin contents of juniper seedlings compared with the control value, whereas abscisic acid content was decreased compared with chemical treatments. Conclusion: The investigated different treatments had an effective role in breaking seed dormancy and improving seedling growth of J. procera, which is facing a notable decline in its population worldwide. Moreover, such an effect was more pronounced in the three phytohormones that succeeded in breaking dormancy and growth of the Juniperus plant than in the other treatments.

BACKGROUND: Temporomandibular joint (TMJ) replacement may be indicated for various pathological conditions, and the type of condition can affect the surgical procedure and outcomes. The causes of limited range of motion after alloplastic TMJ replacement have not been extensively studied. OBJECTIVES: The present study aimed to evaluate the impact of preoperative jaw anatomy and functional status on the immediate and long-term outcomes of total TMJ replacement using a two-component patient-specific TMJ endoprosthesis. MATERIAL AND METHODS: This retrospective study included 31 patients who underwent total TMJ replacement surgery between 2016 and 2020. The main outcome variable was the maximal incisal opening (MIO) after treatment. Secondary outcome variables included MIO improvement and the presence and type of postoperative complications. The primary predictive variable was the preoperative initial MIO. Secondary predictive variables included sex, age, indications for TMJ replacement, preoperative occlusion, condition of the glenoid fossa and/or condyle, shortening of the mandibular ramus, sagittal mandible position, lateral chin deviation, shape of the coronoid process, and type of surgery. RESULTS: The mean preoperative MIO was 13.0 ±8.0 mm, while the mean MIO 1 month after surgery was 20.6 ±5.5 mm, which was not statistically significant. However, at a later follow-up, functional parameters showed a significant improvement (p = 0.003), with a mean MIO of 32.5 ±5.0 mm 3 years after surgery. Statistical analysis indicated that the initial mouth opening is the strongest predictor of long-term functional recovery after TMJ replacement. Postoperative complications occurred in 4 cases (12.9%) following patient-specific endoprosthesis (PSE) placement. CONCLUSIONS: The use of PSEs for TMJ replacement has enabled the restoration of anatomical relationships in complex clinical cases and an improvement in mouth opening. The preoperative MIO was the only factor that significantly influenced long-term functional outcomes.

OBJECTIVES: Bacillus anthracis infection is a worldwide zoonosis that affects the most vulnerable population and has a high mortality rate without treatment, especially in non-cutaneous presentations. Cutaneous scarification is still common in some regions of the world for the treatment of certain diseases as part of traditional medicine. We describe a series of cutaneus anthrax from a rural setting in Angola where cutaneus scarification is common. CASE PRESENTATION: This is a retrospective observational study describing a series of cutaneous anthrax cases from Cubal (Angola), many of whom were treated with skin scarification before admission. A total of 26 cases were diagnosed from January 2010 to December 2018. None of the cases were confirmed and eight (30.8%) were probable cases according to the Centers for the Disease Control and Prevention anthrax case definition. The median age was 11 (4.7-30.5) years, 17 (65.4%) had lesions on the head, face, or neck and 15 (57.7%) were treated with cutaneous scarification. Nine (34.6%) patients died. Traditional cutaneous scarification was significantly associated with cutaneous superinfection, respiratory, systemic involvement, and death. CONCLUSION: Our case series points to increased complications and worse outcome of cutaneous anthrax disease if treated with skin scarification.

The monkeypox epidemic, which became unusually widespread among humans in 2022, has brought awareness about the necessity of smallpox vaccination of patients in the risk groups. The modern smallpox vaccine variants are introduced either intramuscularly or by skin scarification. Intramuscular vaccination cannot elicit an active immune response, since tissues at the vaccination site are immunologically poor. Skin has evolved into an immunologically important organ in mammals; therefore, intradermal delivery of a vaccine can ensure reliable protective immunity. Historically, vaccine inoculation into scarified skin (the s.s. route) was the first immunization method. However, it does not allow accurate vaccine dosing, and high-dose vaccines need to be used to successfully complete this procedure. Intradermal (i.d.) vaccine injection, especially low-dose one, can be an alternative to the s.s. route. This study aimed to compare the s.s. and i.d. smallpox immunization routes in a mouse model when using prototypic second- and fourth-generation low-dose vaccines (104 pfu). Experiments were conducted using BALB/c mice; the LIVP or LIVP-GFP strains of the vaccinia virus (VACV) were administered into the tail skin via the s.s. or i.d. routes. After vaccination (7, 14, 21, 28, 42, and 56 days post inoculation (dpi)), blood samples were collected from the retro-orbital venous sinus; titers of VACV-specific IgM and IgG in the resulting sera were determined by ELISA. Both VACV strains caused more profound antibody production when injected via the i.d. route compared to s.s. inoculation. In order to assess the level of the elicited protective immunity, mice were intranasally infected with a highly lethal dose of the cowpox virus on 62 dpi. The results demonstrated that i.d. injection ensures a stronger protective immunity in mice compared to s.s. inoculation for both VACV variants.

RESUMO

Transient receptor potential (TRP) channels are a class of transmembrane proteins that can sense a variety of physical/chemical stimuli, participate in the pathological processes of various diseases and have attracted increasing attention from researchers. Recent studies have shown that some TRP channels are involved in the development of pathological scarification (PS) and directly participate in PS fibrosis and re-epithelialization or indirectly activate immune cells to release cytokines and neuropeptides, which is subdivided into immune inflammation, fibrosis, pruritus and mechanical forces increased. This review elaborates on the characteristics of TRP channels, the mechanism of PS and how TRP channels mediate the development of PS, summarizes the important role of TRP channels in the different pathogenesis of PS and proposes that therapeutic strategies targeting TRP will be important for the prevention and treatment of PS. TRP channels are expected to become new targets for PS, which will make further breakthroughs and provide potential pharmacological targets and directions for the in-depth study of PS.

High temperatures, soil salinity, a lack of available water, loose soils with reduced water holding, and low soil fertility are obstacles to restoration efforts in degraded drylands and desert ecosystems. Improved soil physical and chemical properties, seed germination and seedling recruitment, and plant growth are all proposed as outcomes of seed enhancement technologies (SETs). Seed priming, seed coating, and seed scarification are three SETs' methods for promoting seed germination and subsequent plant development under unfavorable environmental conditions. Various subtypes can be further classified within these three broad groups. The goals of this review are to (1) develop a general classification of coating and scarification SETs, (2) facilitate the decision-making process to adopt suitable SETs for arid lands environments, and (3) highlight the benefits of coating and scarification SETs in overcoming biotic and abiotic challenges in ecological restoring degraded dryland. For rehabilitating degraded lands and restoring drylands, it is recommended to 1) optimize SETs that have been used effectively for a long time, particularly those associated with seed physiological enhancement and seed microenvironment, 2) integrate coating and scarification to overcome different biotic and abiotic constraints, and 3) apply SET(s) to a mixture of seeds from various species and sizes. However, more research should be conducted on developing SETs for large-scale use to provide the required seed tonnages for dryland restoration.

Production of biofuel from lignocellulosic biomass is relatively low due to the limited knowledge about natural cell wall loosening and cellulolytic processes in plants. Industrial separation of cellulose fiber mass from lignin, its saccharification and alcoholic fermentation is still cost-ineffective and environmentally unfriendly. Assuming that the green transformation is inevitable and that new sources of raw materials for biofuels are needed, we decided to study cell death-a natural process occurring in plants in the context of reducing the recalcitrance of lignocellulose for the production of second-generation bioethanol. "Members of the enzyme families responsible for lysigenous aerenchyma formation were identified during the root hypoxia stress in Arabidopsis thaliana cell death mutants. The cell death regulatory genes, LESION SIMULATING DISEASE 1 (LSD1), PHYTOALEXIN DEFICIENT 4 (PAD4) and ENHANCED DISEASE SUSCEPTIBILITY 1 (EDS1) conditionally regulate the cell wall when suppressed in transgenic aspen. During four years of growth in the field, the following effects were observed: lignin content was reduced, the cellulose fiber polymerization degree increased and the growth itself was unaffected. The wood of transgenic trees was more efficient as a substrate for saccharification, alcoholic fermentation and bioethanol production. The presented results may trigger the development of novel biotechnologies in the biofuel industry.

Persistence in the soil is a function of seed physiology, particularly non-germination and inherent lifespan. However, for seeds with mechanical dormancy, non-germination is also a function of the composition and activity of the soil microbiota. We attempted to screen out microorganisms in the soil that can specifically and rapidly decompose the hard fruit pericarps of Tilia miqueliana Maxim., a unique native tree species in China. Using the classical replica plating method, more than 100 different culturable microorganisms that could rapidly erode the pericarp were collected from the surface of pericarps under different culture conditions. At the same time, we successfully extended the concept of metagenomics and applied it to the identification of mixed artificial cultures. The decomposition process of the pericarps in soil was also simulated artificially. The physical and chemical data suggested a potential mechanism of microbial scarification and cracking in pericarp, whilst the embryos inside the eroded fruits retained good viability. Our discoveries could pave the way for the removal of physical and mechanical obstacles that prevent hard coat seeds from germinating. We anticipate that the use of this technology will improve the germination of other hard coat seeds. More research is needed to investigate the impacts on other seeds. The findings of this research can inform the design of experiments on the seed ecology of persistence.

The purpose of this study was to compare the effectiveness of arthroscopic-assisted lateral pterygoid muscle release versus scarification of retrodiscal tissues in the treatment of temporomandibular joint internal derangement. A prospective, comparative, randomized clinical trial involved 16 patients with TMJ internal derangement. Arthroscopic assisted release of lateral pterygoid muscle was assigned to one group of patients (Group I). Group II patients received arthroscopic assisted scarification of retrodiscal tissues. Data collected through functional examination including visual analogue scale (VAS), maximum mouth opening, lateral excursion, and clicking sound immediately and after 3, 6, and 12 months. Pre- and post-operative MRI was used to assess disc position. The VAS scores decreased in both groups at the end of the follow-up period (0.45 vs 6.75, and 1.13 vs 6.50 in group I and II respectively; P<0.001). The maximum mouth opening improved to 32.9.50 ± 1.69 mm in group I, and 30.49 ± 0.93 in group II (P<0.001). Lateral excursion improved in both groups (P<0.001). Clicking sounds disappeared in all patients. Within the limitations of the study, it seems that arthroscopic assisted release of lateral pterygoid and scarification of retrodiscal tissues may be beneficial in management of anteriorly displaced discs that don't respond to conservative treatments.

BACKGROUND: Vanilla planifolia is the most widely cultivated species of vanilla with high economic importance. However, seed germination under artificial conditions is difficult and yields low germination percentages. The seeds are adapted to endozoochorous dispersal, and we therefore tried to simulate the conditions in the digestive tract by acid scarification of seeds. RESULTS: Immature seeds lacking dormancy, used as a control, showed the highest germination percentage. Among the treatments tested for mature seeds, the hydrochloric acid treatments were significantly the best in breaking dormancy and inducing germination, irrespective of the acid concentration and the presence of pepsin. Conventional treatment with a hypochlorite solution induced much lower germination percentage. Sulphuric acid at concentration 50% was too strong and caused damage to the seeds. Important factor is also high cultivation temperature 30 °C as there was nearly no germination at 25 °C. CONCLUSIONS: Our protocol significantly improves the efficiency of generative propagation of vanilla and allows for significantly higher germination percentages than previously described. The strongly positive effect of hydrochloric acid may be related to the adaptation of seeds to endozoochorous dispersal.

Rainsplash erosion on forested hillslopes can be increased by both wildfires and post-fire salvage logging, especially under semi-arid Mediterranean conditions. However, few studies have compared rainsplash erosion among forest sites impacted by logging to other forest areas. To fill this gap, this study has evaluated surface runoff and soil erosion in a burnt and logged (manually or mechanically) pine forest of Central-Eastern Spain under simulated rainfall and compared it to unlogged and unburnt plots. Compared to the unburnt plots, surface runoff significantly increased (over 150%) in logged areas, with a peak of 220% on the areas directly subjected to logging machinery. Peak runoff was substantially increased by fire (+130%) and less by logging (+8. Soil loss due to rainsplash erosion was about 235% (manual logging) to 750% (mechanical logging) higher compared to the unburnt plots. Wildfire exerted a much higher soil disturbance compared to salvage logging, with a soil hydrological response that can be up to an order of magnitude higher. The increased runoff and erosion rates in response to wildfire and logging were ascribed to soil compaction, which increased on average 60% on logged plots as well as to the removal of vegetation cover (-80%), whereas soil roughness played a minor role. From these results, we suggest using lightweight machinery in burnt soils, to reduce surface runoff and erosion. The possibility of building contour felled log debris using the burnt wood may also be considered, in order to retain the eroded sediments.

Alterations in global climate via extreme precipitation will have broadscale implications on ecosystem functioning. The increased frequency of drought, coupled with heavy, episodic rainfall are likely to generate impacts on biotic and abiotic processes across aquatic and terrestrial ecosystems. Despite the demonstrated shifts in global precipitation, less is known how extreme precipitation interacts with biophysical factors to control future demographic processes, especially those sensitive to climate extremes such as organismal recruitment and survival. We utilized a field-based precipitation manipulation experiment in 0.1 ha forest canopy openings to test future climate scenarios characterized by extreme precipitation on temperate tree seedling survival. The effects of planting seedbeds (undisturbed leaf litter/organic material vs. scarified, exposed mineral soils), seedling ontogeny, species, and functional traits were examined against four statistically defined precipitation scenarios. Results indicated that seedlings grown within precipitation treatments characterized by heavy, episodic rainfall preceded by prolonged drying responded similarly to drought treatments lacking episodic inputs. Moreover, among all treatment conditions tested, scarified seedbeds most strongly affected seedling survivorship (odds ratio 6.9). Compared with any precipitation treatment, the effect size (predicted probabilities) of the seedbed was more than twice as important in controlling seedling survivorship. However, the interaction between precipitation and seedbed resulted in a 27.9% improvement in survivorship for moisture-sensitive species. Seedling sensitivity to moisture was variable among species, and most closely linked with functional traits such as seed mass. For instance, under dry moisture regimes, survivorship increased linearly with seed mass (log transformed; adjusted R2 = 0.72, p < 0.001), yet no relationship was apparent under wet moisture regimes. Although precipitation influenced survival, extreme rainfall events were not enough to offset moisture deficits nor provide a rescue effect under drought conditions. The relationships reported here highlight the importance of plant seedbeds and species (e.g., functional traits) as edaphic and biotic controls that modify the influence of extreme future precipitation on seedling survival in temperate forests. Finally, we demonstrated the biophysical factors that were most influential to early forest development and that may override the negative effects of increasingly variable precipitation. This work contributes to refinements of species distribution models and can inform reforestation strategies intended to maintain biodiversity and ecosystem function under increasing climate extremes.

Trafficking in human beings is an ancient phenomenon. The fight against trafficking in human beings is conceived as a priority by the European Union. Trafficked victims experience many types of abuse and neglect. Indeed, all human beings hold the right to have an identity and an estimated age as an assertion of their existence in the society, as expressly stated in the U.N. Convention on the Rights of the Child. Italy has been the scene of this phenomenon for many years. Since identification represents a fundamental human right and a prerequisite for any other measure of assistance and protection, the Italian procedure provides the execution of an interview and a forensic examination. This process takes time and requires willingness to listen and to create a trusting doctor-patient relationship. Although skin lesions in trafficking victims may be due to torture or other forms of mistreatment or abuse, they may also be related to ethnic practices. Here we demonstrate the importance of conducting a structured interview along with an accurate forensic examination to correctly discriminate the origin of skin lesions in trafficking victims.

Accelerated global warming is leading to the loss of plant species diversity, and ex situ preservation of seeds is becoming an increasingly important aspect of species conservation. However, information on dormancy and germination is lacking in many endangered species. Amsonia elliptica (Apocynaceae) is the only Amsonia species native to Korea, and the South Korean Ministry of Environment has designated it Class II endangered wildlife. Nevertheless, the dormancy class and the dormancy breaking method for seeds of this species for germination are not precisely known. We identified the structure of A. elliptica seeds and the causes of dormancy, which inhibits germination. In addition, we tried to develop an effective germination promotion method by testing the wet stratified condition, which breaks dormancy, and the form of gibberellin that can replace it. Fresh seeds of A. elliptica imbibe water, but the covering layers (endosperm and seed coat) inhibit germination by mechanically restricting the embryo. Initial germination tests confirmed low embryo growth potential and physiological dormancy (PD). Restriction due to the covering layer was eliminated by seed scarification, and abnormal germination was observed. After 12 weeks of cold moist stratification at 4°C, only 12% of seeds germinated. However, 68.8% of seeds subjected to 8 weeks of warm moist stratification followed by 12 weeks of cold stratification germinated, indicating that warm stratification pretreatment before cold stratification is effective in breaking dormancy. A. elliptica seeds exhibited intermediate PD. Furthermore, 61.3% of seeds soaked in 500 mg/L GA4+7 for 14 days and incubated at 25/15°C germinated. Therefore, GA4+7 rapidly broke the dormancy of A. elliptica seeds compared with warm plus cold stratification treatment, thus providing an efficient method for seedling production.

Tamaulipan thornforests in south Texas and northeast Mexico are a conservation hotspot. Shortages of native seedlings limit regional restoration and are largely driven by knowledge gaps regarding propagation of the 75+ thornforest species planted during restorations. We previously investigated three thornforest species with low or inconsistent germination or seedling survival: Ebenopsis ebano (Fabaceae), Cordia boissieri (Boraginaceae), and Zanthoxylum fagara (Rutaceae), and identified the types and dosages of chemical seed treatments that maximized germination. However, chemical treatments were performed in isolation and combinational treatments may be required to break dormancy or maximize germination. This study builds on prior work by investigating the effects of all possible combinations of sulfuric acid (SA), gibberellic acid (GA), and indole-3-butyric acid (IBA) treatments on germination of the same focal species, and further quantified the combined effects of five chemical treatments, three stratification treatments, and six soil mixture types on the germination and seedling performance of the focal species. Ebenopsis ebano germination peaked with SA and was not improved with additional chemical treatments. Cordia boissieri germination was highest with GA only in our indoor experiment but peaked with GA + IBA + SA in our outdoor experiment. Zanthoxylum fagara germination was near zero in all treatments. Stratification treatments marginally reduced E. ebano germination and reduced C. boissieri seedling height. Soil type had significant impacts on E. ebano germination and leaf abundance (residual differences up to 40% or 4 leaves, respectively) and influenced some of the effects of chemical treatments. These results enhance our understanding of thornforest seed ecology and best practices for nursery propagation of seedlings.

The skin is the site of host invasion by the mosquito-borne Plasmodium parasite, which caused an estimated 229 million infections and 409,000 deaths in 2019 according to WHO World Malaria report 2020. In our previous studies, we have shown that skin scarification (SS) with a P. falciparum circumsporozoite (CS) peptide in the oil-in-water adjuvant AddaVax containing a combination of TLR 7/8 and TLR 9 agonists can elicit sporozoite neutralizing antibodies. SS with AddaVax + TLR agonists, but not AddaVax alone, elicited CD4+ Th1 cells and IgG2a/c anti-repeat antibody. To explore the innate immune responses that may contribute to development of adaptive immunity following SS, we examined the skin at 4h and 24h post priming with CS peptide in AddaVax with or without TLR agonists. H&E stained and IHC-labeled dorsal skin sections obtained 24h post SS demonstrated a marked difference in the pattern of infiltration with F4/80+, CD11b+ and Ly6G+ cells at the immunization site, with the lowest intensity noted following SS with AddaVax + TLR agonists. Serum collected at 4h post SS, had reproducible increases in IL-6, MIP-3α, IL-22 and IP-10 (CXCL10) following SS with AddaVax + TLR agonists, but not with AddaVax alone. To begin to decipher the complex roles of these pro-inflammatory cytokines/chemokines, we utilized IP-10 deficient (IP-10 -/-) mice to examine the role of this chemokine in the development of anti-repeat antibody response following SS. In the absence of IP-10, the levels of Th1-type IgG2a/c antibody and kinetics of the primary anti-repeat antibody response were reduced following prime and boost. The IP-10 chemokine, present as early as 4h post prime, may provide an early serological marker for rapid screening of adjuvant formulations and delivery platforms to optimize SS-induced humoral immunity to CS repeats as well as other pathogens.

The herbaceous perennial legume Mimosa pudica is an invasive weed in many tropical and subtropical regions and a serious problem for farmers since it is difficult to eliminate from crop field by hand. Moreover, it has water impermeable seeds, i.e., physical dormancy (PY), which could persist in the soil seed bank for a long period of time, thus making it a big challenge to control. The aims of this study were to test the effect of various laboratory methods on breaking PY in seeds of M. pudica, to identify the site(s) of water entry into seeds of M. pudica and compare results of dormancy-breaking methods for seeds of M. pudica with those of 36 other species of Mimosa reported in the literature. Mechanical scarification, wet heat and cycles of wet heat and ice water effectively broke PY in seeds of M. pudica. Following wet heat at 80 °C for 10 min, water uptake was via the hilar region but not the pleurogram; small cracks made in the pleurogram by this treatment were not deep enough for water to enter the seed. Neither tolerance to summer temperatures nor PY is likely to be the cause of invasiveness of this species, since seeds of rare and endemic species of Mimosa also tolerate summer temperatures and have PY.

RESUMO

The effect of the main fire factors (smoke, ash, charcoal and heat) can influence the germination of species through their seeds. Hence, a methodology has been devised in order to have a common protocol for those who work in this area and serve as a valuable tool to compare different species that can be beneficial or detrimental to the conditions of a forest fire. Moreover, the methodology was completed with a check of the viability of the seeds before starting the germination process and another viability test with the non-germinated seeds (dormant or dead) after carrying out the germination test. In this way, the study of the main factors of fire, in combination with the study of the viability of the seeds, can be very useful to know the reproductive behaviour of herbaceous, shrub and tree species in a forest fire scenario. In addition, from the data obtained during the germination process of the seeds, a series of explained parameters can be obtained to better interpret the data acquired from the laboratory experiment and subsequent comparison. For all these reasons, the study of the germination of species in laboratory conditions can help us understand how they behave in the field.

<b>Background and Objective:</b> Intensive and commercial development of areca nut requires the provision of high-vigour areca nut. This study aimed to evaluate the effect of scarification and seeds biomatriconditioning using endophytic-rhizobacteria in increasing seedlings vigor of areca nut. <b>Materials and Methods:</b> The research was carried out at the Agronomy Unit of Agrotechnology Laboratory, Agriculture Faculty, Halu Oleo University, from November, 2020-March, 2021. The research design was split-plot in a Completely Randomized Design (CRD). The main plot, seed scarification, consisted of 2 treatments, without scarification and scarification. Sub-plots, seeds biomatriconditioning using endophytic-rhizobacteria, consisted of 6 treatments, control, L1-R, M5-R, LA6-R, LA2-E and RJ6-R. <b>Results:</b> The results showed that the scarification treatment did not affect the seedlings vigor of the areca nut. Seed biomatriconditioning using endophytic-rhizobacteria was able to increase seedling vigor both without scarification and with scarification. There were 3 isolates of endophytic-rhizobacteria which were more able to increase the vigor of areca nut without scarification, namely L1-R, LA6-R and LA2-E, with an increase of 137, 104 and 102%, respectively compared to the control, while in scarified seeds, L1-R isolate was able to increase the seedlings vigor of areca nut by 194% compared to the control. <b>Conclusion:</b> Scarification treatment did not affect the vigor of the areca nut. Seed biomatriconditioning with endophytic-rhizobacteria was able to increase the vigor of areca nut seeds either without scarification or with scarification.

Pleural injury and subsequent loculation is characterized by acute injury, sustained inflammation and, when severe, pathologic tissue reorganization. While fibrin deposition is a normal part of the injury response, disordered fibrin turnover can promote pleural loculation and, when unresolved, fibrosis of the affected area. Within this review, we present a brief discussion of the current IPFT therapies, including scuPA, for the treatment of pathologic fibrin deposition and empyema. We also discuss endogenously expressed PAI-1 and how it may affect the efficacy of IPFT therapies. We further delineate the role of pleural mesothelial cells in the progression of pleural injury and subsequent pleural remodeling resulting from matrix deposition. We also describe how pleural mesothelial cells promote pleural fibrosis as myofibroblasts via mesomesenchymal transition. Finally, we discuss novel therapeutic targets which focus on blocking and/or reversing the myofibroblast differentiation of pleural mesothelial cells for the treatment of pleural fibrosis.
